چکیده

Soluble enzyme preparations from irradiated cell suspension cultures of parsley (Petroselinum hortense Hoffm.) catalyse the conversion of flavanone to flavone, dihydroflavonol and flavonol. These reactions require 2-oxoglutarate, Fe2+ and ascorbate as cofactors. In the presence of these cofactors conversion of dihydroflavonol to flavonol was also observed. With this system in vitro biosynthesis of radioactive flavone, dihydroflavonol and flavonol from [2-14C]malonyl-CoA and 4-coumaroyl-CoA in good yield and with high specific activity is possible. We postulate that synthesis of flavone and flavonol from flavanone proceeds via 2-hydroxyand 2,3-dihydroxyflavanone, respectively, with subsequent dehydration. The microsomal fraction of the parsley cells contains an NADPH-dependent flavanone 3'hydroxylase.

Enzymes of General Phenylpropanoid Metabolism and of Flavonoid Glycoside Biosynthesis in Parsley: Differential Inducibility by Light during the Growth of Cell Suspension Cultures.

Several enzymes of phenylpropanoid metabolism showed large changes in their inducibility by light during the growth cycle of cell suspension cultures from parsley (Petroselinum hortense Hoffm.). Two of the three enzymes of general phenylpropanoid metabolism (group I) and six of the approximately 13 enzymes of the flavone and flavonol glycoside pathways (group II) were investigated. Both enzymes...

Flavonol Synthase Activity and the Regulation of Flavonol and Anthocyanin Biosynthesis during Flower Development in Dianthus caryophyllus L. (Carnation)

Flavonol synthase (FLS) was demonstrated in crude extracts from flower buds o f Dianthus caryophyllus (carnation). The enzyme catalyzed the conversion o f dihydrokaempferol and dihydroquercetin to kaempferol and quercetin, respectively. The reaction required 2-oxoglutarate, ferrous ion and ascorbate as co-factors and had a pH optimum at about 7.4. The demonstration o f FLS activity allowed comp...

13C-NMR, Apigenin, Kaempferol, Flavonoid, Biosynthesis Cell suspension cultures of parsley (Petroselinum hortense) were incubated with [l,2-13C2]acetate during a period of active flavonoid production. The flavonoid glycosides were extracted with ethanol, hydrolyzed in dilute acid, and the aglycones purified by chromatography. Apigenin, a flavone, and kaempferol, a flavonol, were analyzed at 67....

Induction and Characterization of a NADPH-Dependent Flavone Synthase from Cell Cultures of Soybean

Microsomal preparations from osmotically stressed soybean cells catalyze the conversion of (25)-naringenin to apigenin in presence of N A D P H . In contrast, such preparations from normal soybean cells or from elicitor-challenged cells catalyze the conversion o f (25)-naringenin to genistein (isoflavone synthase). It is concluded that osm otic stress of the cells causes a switch from isoflavon...

High-flavonol tomatoes resulting from the heterologous expression of the maize transcription factor genes LC and C1.

Flavonoids are a group of polyphenolic plant secondary metabolites important for plant biology and human nutrition. In particular flavonols are potent antioxidants, and their dietary intake is correlated with a reduced risk of cardiovascular diseases. Tomato fruit contain only in their peel small amounts of flavonoids, mainly naringenin chalcone and the flavonol rutin, a quercetin glycoside. To...
